I tried making it but it doesnt work,it still msgs me and my friend even if only one enters:

on region enter:
wait 2 ticks
if "%region at player%" contains "1v1":
if {playersinregion} is 2:
make console execute command "/msg swordenn works"
make console execute command "/msg shap1n works"

Your {playersinregion} isn't being incremented when a player enters, so it may not reflect the actual count.

The else if: statement is incomplete and needs a proper condition.

on region enter:
    wait 2 ticks

        add player to {playersinregion::*}
        if size of {playersinregion::*} is 2:
            make console execute command "/msg swordenn works"
            make console execute command "/msg shap1n works"
